Abstract

Spermatozoa sapi Bali rentan terhadap kerusakan akibat stres oksidatif selama kriopreservasi, yang dapat menurunkan kualitas semen beku. Penelitian ini bertujuan mengevaluasi pengaruh penambahan antioksidan alami (ekstrak teh hijau) dan sintetis (glutathione) pada pengencer Cauda Epididymal Plasma (CEP) terhadap kualitas semen beku sapi Bali. Rancangan percobaan yang digunakan adalah Rancangan Acak Lengkap (RAL) dengan tiga perlakuan dan enam ulangan, pengencer CEP dasar (kontrol), CEP + 0.75 mM glutathione, dan CEP + 0.15% ekstrak teh hijau. Evaluasi kualitas spermatozoa dilakukan saat before freezing dan post thawing, meliputi motilitas menggunakan Computer-Assisted Semen Analysis (CASA), viabilitas menggunakan pewarna eosin-nigrosin dan integritas membran dan uji Hypo-Osmotic Swelling Test (HOST). Hasil penelitian menunjukkan bahwa penambahan antioksidan secara signifikan meningkatkan kualitas spermatozoa dibanding dengan kontrol (p<0,05). CEP yang ditambahkan 0,15% ekstrak teh hijau memberikan hasil terbaik dalam mempertahankan post-thawing motilitas (49,2 ± 2%), viabilitas (67,4 ± 2,6%), dan integritas membran (62,1 ± 1,9%). Kesimpulannya, ekstrak teh hijau dan glutathione secara signifikan lebih efektif dibandingkan kontrol, dengan ekstrak teh hijau cenderung memberikan nilai tertinggi pada motilitas, viabilitas, dan integritas membran post-thawing.
